Epitope mapping of mAb806
The mAb 806 is a monoclonal antibody which acts againsts a mutant form of the EGF receptor
variously called DeltaEGFR, de2-7 EGFR, or EGFRvIII. This receptor has been observed to be
frequently highly expressed in human malignant gliomas and has been reported for cancers of
the lung, breast, and prostate. The antibody does not bind to wild type EGF receptor and
possesses significant antitumor activity. To gain a better idea of the mechanism of action of this
antibody, we are attempting to determine the epitope of binding of mAb 806. Preliminary results
indicate that the epitope is away from the novel splice site in the protein which is generated as
a consequence of the mutation.
